What is Azisign-500 by Hab Pharmaceuticals and Research Ltd.?

Few antibiotics can treat an infection with a single dose or with just three tablets. Azithromycin can, and Azisign-500 delivers it as a full-strength adult tablet: 500 mg per pill, five pills per pack, made by Hab Pharmaceuticals and Research Ltd.

The substance is a macrolide (azalide) antibiotic. Inside the bacterium it blocks the exit tunnel of the ribosome, where new proteins are made, so the cell cannot produce the enzymes and structural proteins it needs. Azithromycin has a very large volume of distribution: most of each dose ends up in cells and tissues rather than in the blood, and it is released from there slowly, with an elimination half-life of a few days.

Main indications

Azithromycin is chosen for infections where its spectrum fits and local resistance is low:

  • uncomplicated bacterial infections of the skin and soft tissue;
  • early Lyme disease with the typical expanding red rash, as an option when first-line drugs cannot be used;
  • respiratory infections, from sinusitis to atypical pneumonia;
  • chlamydia and other genital infections;
  • severe travelers' diarrhea in regions where other antibiotics no longer work.

It is not a first-choice acne treatment and has no effect on viral infections, fungal skin infections or allergic rashes.

How Azisign-500 is dosed

Typical adult regimens with this strength are one 500 mg tablet once a day for three days, or a single 1 g dose (two tablets taken together) for chlamydia and for some cases of travelers' diarrhea. For skin infections the three-day course is the usual fit for this strength; the five-day scheme with 250 mg on days two to five needs a smaller tablet. A pack of five tablets covers one three-day course with two tablets to spare; it is not meant to be taken as five days of 500 mg unless a doctor prescribes that.

Tablets can be taken with food if they upset the stomach. Dose and duration must be agreed with a qualified medical professional, especially for children, older people and anyone with heart, liver or kidney disease.

Side effects and who should avoid it

The most likely problems are diarrhea, nausea and stomach cramps. Taste changes, headache and a passing rash are possible. Hearing loss or ringing in the ears has been reported, mostly with long courses. The rare but serious effects are a prolonged QT interval with risk of arrhythmia, liver injury, severe skin reactions and Clostridioides difficile colitis.

Azithromycin should be avoided by people allergic to any macrolide and used with care with heart rhythm disorders, a slow pulse, low potassium, or drugs such as some antiarrhythmics and antipsychotics that also lengthen the QT interval.
